We have previously reported on the feasibility of weekly topotecan as single-agent therapy in previously treated patients with ovarian cancer.
The large amount of work undertaken by the Alice Springs Desert Park in recent years has considerably advanced our knowledge of the cultivation of many local species. Nevertheless, significant research effort will be required before the full potential of many useful and attractive central Australian genera and species can be fulfilled. In addition to the trade in seed, there may well be a niche opportunity to use the seed to propagate live plants for sale in tubes or pots to the nursery trade. The Alice Springs Desert Park has undertaken some work in this area, producing modest numbers of many local species for wholesale nursery sale Mark Richardson, former botanical curator, Alice Springs Desert Park, pers. comm., 1999 ; , and both Greening Australia and Tangentyere Nursery propagate local species for sale in Alice Springs. While it is too early to adequately assess the longterm viability or potential value of this trade all of the work to date has been subsidised from various sources there may well be a modest but reliable future for this enterprise, and opportunities for Aboriginal people within it.

The clinical success of the water-soluble camptothecins topotecan and irinotecan has stimulated efforts to optimize the efficacy of this class of agents. In an attempt to synthesize new analogues, the chemical structure of camptothecin has been extensively dissected, and structure-activity relationship studies have revealed critical aspects for drug activity 2 ; . Among these, the structure of the lactone E ring and its 20 S ; -hydroxyl configuration are considered essential for the antitumor action 1, 3, 4 ; . The a-hydroxylactone E ring is unstable in water solution at physiologic pH, and camptothecins yield two molecular species, the lactone and the ring-opened carboxylate forms. The opened form is almost inactive and substantially less potent than the lactone form 5, 6 ; . At physiologic pH, the equilibrium between the two species is shifted toward the carboxylate form. The equilibrium is also dependent on the presence of specific binding proteins i.e., human serum albumin, which binds the opened ring shifting the equilibrium towards the carboxylate form; refs. 5, 7 ; . Chemical modifications of the lactone ring, such as transformation into lactam ring, reduction removal of the carbonyl oxygen, or substitution of the 20 S ; -hydroxyl for hydrogen, inactivate the molecule 6, 8, 9 ; , thus implicating the presence of an intact 20-hydroxy lactone ring as an essential requirement for the cytotoxic activity. However, some natural glycoside analogues are characterized by a ring-opened form 10 ; . In particular, one natural constituent of Nothapodytes foetida, named Foetidin I, is a di-coumaroyl spermidine ester of the carboxylate form of camptothecin. The biological significance of these camptothecinlike molecules is still unknown. Among the ways thus far devised to improve the efficacy of DNA-interactive agents, conjugation of a cytotoxic drug to natural polyamines seems an attractive strategy. The polyammonium cations have proved to favor the interaction with the negatively charged phosphates of the DNA backbone maintaining a high degree of translational freedom along the polyanion backbone, thus conferring a high DNA affinity without modifying the capability of the drug to locate appropriate sites of DNA 11 ; . | Topotecan and cisplatinIn summary, the topotecan platinum combination in front-line therapy shows promising activity. The major DLT was schedule-dependent myelosuppression, with greater hematologic toxicity observed when platinum was administered on day 1 of each course. Identifying optimal dosing and a schedule that will maximize the antitumor activity of the topotecan-based combination but limit the severity of myelosuppression requires further investigation. Triplet Regimen The rationale for integrating topotecan into the existing standard as part of a triplet regimen is based on in vitro synergy among topotecan, platinum, and paclitaxel [15] and on non-cross-resistance between topotecan and paclitaxel in clinical trials [27]. As summarized in Table 2, clinical investigations have recently evaluated the antitumor activity and tolerability of topotecan administered as part of a triplet regimen with paclitaxel and platinum [32-34] and Engelholm and Scarfone, personal communications ; . Frasci and colleagues [32] conducted a phase I study to determine the MTD of topotecan given weekly over 30 minutes in combination with cisplatin and paclitaxel, with G-CSF support. Nineteen ovarian cancer patients, 11 of whom were chemotherapy nave, received a combination of cisplatin, 40 mg m2, paclitaxel, 85 mg m2 1-hour infusion ; , and escalating doses of topotecan beginning at 0.75 mg m2 over 30 minutes ; weekly. Of nine chemotherapy-nave ovarian cancer patients who were evaluable for tumor response, seven 78% ; achieved responses, including three CCRs and four PRs. The most frequent serious toxicity was hematologic. A weekly topotecan dose of 2.25 mg m2 was recommended for phase II studies and trandolapril.

Despite this equality, however, a striking dissociation was evident in subsequent inference ability: subjects tested shortly after training showed no evidence of inferential ability, displaying chance performance. Yet those tested following delay periods spanning 12 and 24 hr demonstrated a highly significant facilitation of inference. Therefore, somewhere between 20 min and 12 hr, a process of offline binding of the premise pairs developed, resulting in the increased capability for relational judgments. If, as we claim, inference can take time to develop, then why have previous studies described the ability for inference immediately after training, without the need for such offline delays 57, 20, 21 ; ? Common among these past studies, and distinct from our paradigm, is that participants were trained to ceiling levels on the premise pairs. Notably, in one of the few studies where premise pair knowledge was not at a ceiling, using a similar training criterion to the one we report here, healthy participants were not able to express inference ability when tested immediately after training 8 ; . A further difference also pertains to the structure of training that most paradigms use, often presenting the premise pairs in ascending or descending order during initial learning, a method that may facilitate hierarchical knowledge more quickly, relative to the randomized order used here. Collectively, these prior findings suggest that saturating subjects with premise pair training, or training the pairs in order, can lead to inference without the requisite offline processing. However, in the more ecologically valid circumstance, where individual item knowledge here, the premise pairs ; is not learned in an ordered manner nor overtrained to perfection, time, and especially sleep, appear capable of establishing this relational ability. One potential candidate mechanism of this offline phenomenon is the process of consolidation 22 ; . Numerous studies investigating the consolidation of both declarative and nondeclarative memory have demonstrated the continued modulation of recently acquired information offline 912, 17, 18 ; . Here, we extend the knowledge that consolidation processes benefit individual memory items by demonstrating that similar offline delays can also lead to the associative integration of these item elements into a ``metamemory representation, '' from which can emerge flexible behavioral repertoires, such as inferential judgments.
