Manual placement of the VOI's over different brain regions is prone to subjectivity. Automated registration, semi-quantitative "volume of interest" VOI ; analyses and a comparison of the obtained results to normal values mean and standard deviation ; result in a more reliable and reproducible analysis of brain perfusion images, as compared to manual registration and visual interpretation. The aim of the study was to explore the value of automatic registration to a reference template and to include a predefined VOI map on canine brain perfusion data, using the methodology of the BRASS program from Nuclear Diagnostics. A template was created for both FBP and OSEM reconstructed data. Materials and methods: A 99mTc-ECD image template was created including 12 normal dogs 6 males and 6 females ; , aged between 12-84 months mean: 49 months, SD: 29 ; . Data were acquired for one main energy window and two scatter windows, with a Toshiba GCA9300 triple headed gamma camera, equipped with low energy, high resolution fan-beam collimators. All data were filtered, scatter corrected, re-binned to parallel data and reconstructed with filtered back-projection. Uniform Sorensen attenuation was applied. Each reconstructed image was manually adjusted to one arbitrary chosen image by two experienced persons. A first mean image was calculated from all fitted images. Twelve brain regions were defined on this image, creating a region map. For each registered image the average counts per voxel and the average counts per voxel over all predefined regions were calculated. Regional data were normalised to data from all predefined regions. All manually registered images were then re-registered on the mean image with an automatic registering algorithm, allowing shift, scaling and rotation in all three directions. The automatic registration procedure is based on the downhill Simplex minimization algorithm, using a count difference cost function, tri-linear interpolation and a mask excluding extra-cerebral activity. The same methodology was applied to OSEM reconstructed data. Results: No visual discrepancies were noted after the manual and the automatical fitting procedure, although the standard deviation between the individual cerebral regions regions was highest after manual fit compared to the automatic registration. Conclusions: Automatic fit is feasible for canine brain flow data and has the advantage that semi-quantification can be performed more accurate and less subjectively. It can provide a mean to compensate for size and shape differences between different breeds, a major concern in canine brain imaging.
