Our memories of her capabilities were fully rewarded, and we had a very good turnout! At our May 23 meeting, we listened to presentations by Dave DeGregorio and Buz Crain, two of our members. Prandoni" * randomized 170 patients with venographically confirmed proximal deep vein thrombosis to either Fraxiparin using a weight adjusted regimen of between 12, 500 and 17, 000 anti-Xa IC U twice daily ; or SH administered by continuous IV infusion adjusted to maintain the APTT at 1.5 to 2 times control. The outcome measures were symptomatic recurrence at 10 days and 6 months. At 10 days, 4 of 85 4.7% ; patients receiving SH developed recurrence compared with 1of 85 1.2% ; receiving LMWH. At 6 months, 11 of 85 12.9% ; of SH patients developed recurrent thromboembolism compared with 6 of 85 LMWH patients 7.1% ; , P .2. All patients underwent repeat venography after 7 days of therapy. In the SH patients, the second venogram was improved in 36 patients, unchanged in 35, and worse in 14 patients. In contrast, in the LMWH group the second venogram was improved in 50 patients, unchanged in 28, and worse in 5 patients, P .3. Bleeding occurred in 10.6% of the SH patients compared with 3.5% in the LMWH patients, P .l. The mortality at 6 months was 12 of 85 14.1% ; in the SH group and 6 of 85 7.1% ; in the LMWH group not significant ; . Most deaths were cancer related. Hull et reported the results of their double blind randomized study comparing Logiparin 175 anti-factor Xa U kg daily sc with SH in patients with proximal vein thrombosis. The LMWH was administered in a fixed dose and the SH by continuous infusion adjusted to maintain the APTT in the therapeutic range. The main outcome measure was symptomatic recurrence at 3 months. The incidence of recurrent venous thromboembolism was 6.1% in the SH group and 3.3% in the LMWH group, P .3. The mortality at 3 months was 9.9% in the LMWH group and 21% in the SH group P .04 most deaths were cancer related. At 10 days there were 16 bleeding events in the SH group 10 major ; and seven in the LMWH group one major ; . The difference in major bleeding was significantly greater in the SH group P .05 ; . The results of these studies suggest that in patients with proximal vein thrombosis LMWHs administered in either a fixed or weight adjusted dose sc are more effective and and colace.

Audio Fingerprints AFP's ; are compact, content-based representations of audio signals used to measure distances among them. An AFP has to be small, fast computed and robust to signal degradations. In this paper an entropy based AFP is presented that performed very well when the signal was corrupted with lossy compression, scaling and even 1KHz Lowpass filtering in the experiments. The AFP is determined by computing the instantaneous amount of information of the audio signal in two-second frames with fifty percent overlapping, the resulting entropy signal is binary coded in order to compare different interpretations e.g. live vs. studio recording ; of the same song with good results. The AFP's robustness is compared with that of Haitsma-Kalker's Hash string based AFP with encouraging results.
Non-liver-specific HSS B sub-site, which co-localized with the PPARE 18, 49 ; and is similar to the AF1 site in the GRU 13 ; , was termed the distal AF1 dAF1 ; site. Electrophoresis Mobility Shift Assay--To begin characterizing the patterns of hepatic nuclear proteins that bind to the dAF1 and dAF2 sites, we performed mobility shift assays. A similar pattern of binding between the AF1 and dAF1 sites and between the AF2 and dAF2 sites of the PEPCK-C gene promoter to nuclear proteins was noted using electrophoresis mobility shift assay. There was an efficient competition obtained only between the analogous sites of each couple. Thus, the band shift of the dAF1 probe was equally competed by the same amount of nonradioactive self-competitor dAF1 site ; or its partner AF1 site ; , whereas neither the dAF1 nor AF1 sites competed with binding to the dAF2 probe. Likewise, although the band shift of the dAF2 probe was efficiently competed by itself and by the AF2 sequence, neither dAF2 nor AF2 competed with the band shift of the dAF1 probe Fig. 3, a and b, respectively ; . The similarity in protein binding between the dAF2 and AF2 sites implies that dAF2 site likely constitutes a binding recognition site for the HNF-3 Foxa2 ; and FOXO1, members of the Forkhead gene family, as found for the AF2 site 50, 51 ; 23 ; . The similarity between dAF1 and AF1 also suggests that non-steroid nuclear receptors bind to dAF1, as previously established for AF1 51 ; . To assess the identity of hepatic proteins binding to dAF1 and dAF2, gel shift analysis was performed using specific antibodies. The dAF1 probe was supershifted by the addition of an antibody to HNF-4 to an extent that corresponded to its abundance in hepatic nuclei 52 ; Fig. 3a ; . However, the antibody to PPAR did not affect the binding of proteins to the dAF1 site. Binding of members of the Forkhead family of transcription factors was demonstrated using the dAF2 site as a probe Fig. 3b ; . The results showed diminished intensity of the bound lower band relative to the upper bound band ; , with antibodies against and colesevelam.

Here, we find that the suppression of the vaccine response apparently is due at least in part to the NKT cell, because we can enhance vaccine efficacy by blockade of IL-13 or use of CD1-deficient mice that lack NKT cells. Granulocyte macrophage colony-stimulating factor GMCSF ; alone has been used as an adjuvant in several vaccine studies 2023 ; . In previous work, we have shown that GM-CSF enhances the magnitude of both cellular and humoral immune responses to vaccine constructs by recruiting professional antigen-presenting cells APCs ; to inductive sites without skewing Th phenotype 24, 25 ; . We have also found that CD4 Th1 cell responses favor CTL induction and that GM-CSF acts synergistically with the Th1-inducing cytokine IL-12 to elicit CTL 24, 26 ; . However, in a CD4 T cell-mediated viral protection model a third cytokine, tumor necrosis factor , was necessary to achieve complete protection 25 ; . We have recently established that levels of CD40L expression on activated CD4 Th cells correlated with functional ability to provide help for CTL induction 27 ; , and we as well as others have shown that the mechanism of help involves signals communicated through CD40LCD40 interaction to activate professional APCs to produce IL-12, IL-15, and enhanced costimulation 12, 2731 ; . An expression plasmid for CD40L has been used as an adjuvant in a DNA vaccine to increase cellular and humoral immunity 32, 33 ; , and it has recently been shown that tumor cells transduced with the genes for GM-CSF and CD40L are effective at recruiting tumor-infiltrating dendritic cells capable of eliciting cellular immunity by a cross-priming mechanism 34 ; . However, such a combination of GM-CSF and CD40L has not been tested for synergy in a vaccine. Therefore, we tested the combination of GM-CSF and CD40L for the ability to enhance synergistically vaccine induction of CTL responses. We reasoned that if GMCSF recruits more professional APCs to the draining lymph nodes 25 ; , and then CD40L matures and activates them, the combination might be synergistic. Further, if the primary role of T help is to activate APCs through GM-CSF and CD40L, we asked whether the combination of these might substitute for CD4 T cell help. Here, we find that this combination is not and comfrey.
